I'm happy with most of my lens line-up: 100-400L, 70-200 F/4 L IS, 50mm f/1.4 and 16-35L but...
I kinda get the hankering for a fisheye lens (which would take me beyond the 16mm end of the 16-35) and I could justify upgrading my macro lens (which also gets pressed into use as a portrait lens) from 105mm Sigma to 100 Canon. I also have a non-Canon p&s and whilst it's very good and does what I want it to as a pocket camera, it doesn't excite me the way that a DSLR does... so it might go.
My bodies are 5D and 1DMkII. Neither lens "upgrade" is essential and I imagine that both might happen over the fullness of time but if you were me which would you go for first and why?
