Yes
My Time Machine backup drive has failed and I need to replace it.

I normally just use a USB drive to do this.

Does anyone know if I can partition a 500GB USB drive and use 320GB for Time Machine and the rest to backup another USB drive manually ?

Yep, peice of cake. I do exactly what the guy in the article describes with one partition for TM and another partition for general file storage, all shared over the network via my Airport Express.
All I need now is for Aperture to be able to support updating vaults on network disks.

Just remember that Time Machine backs up until the partition is full then deletes your oldest backups so the bigger the partition the further back you can go.

I have a 1.5TB drive partioned just as you suggest to 1TB & 500MB which is used as a scratch disk for Photoshop.
