Personally, I would defrag it if you copy a lot of data to & from it regularly. If you're just copying data to it for backup then there's probably no need
Like all drives if its heavily used it will benefit from defragging if its becoming slow. If you dont know what defragging is - its basically a reorganised of all the data so that all the bits of the file are located together to make reading it quicker.
