Whats the best way to highlight and help the eyes pop, is it mainly down PP or is something that you need to do whilst taking the shot ie flashes etc?

I have CS5 and LR although not a pro in either but am capable of finding my way around.

A nice catchlight helps. You can then boost eye contrast and sharpness in post.
 
Two ways I use.

Out of camera I might bounce my flash at 105 degrees and use at the back of it the little popup white card which deflect some light forward. That give a really nice pop to the eyes.

Another technique I use is in Aperture. I select a small brush with edge detection and brush sharpening in just over the eye. Go around the eye lashes and through the eye. Then depending on how it is, zoom in a little and apply a brush to lighten the eye white a touch.

Then zoom out and see how it looks compared to the rest and tone down the settings to moderate it perhaps and make it look natural.
